Target description
Integrin beta1 (UniProt: P05556; also known as Fibronectin receptor subunit beta, Glycoprotein IIa, GPIIA, VLA-4 subunit beta, CD29) is encoded by the ITGB1 (also known as FNRB, MDF2, MSK12) gene (Gene ID: 3688) in human. Integrins are heterodimeric cell surface receptors composed of a variable alpha subunit and a conserved beta subunit. The exact combination of various alpha and beta subunits dictates the binding specificity of integrins to different ECM components. Integrins are not constitutively active. Activation of an integrin from low ligand-binding affinity state to high ligand-binding affinity state requires conformational change that can originate from either from their cytoplasmic or extracellular interactions. Integrin beta-1 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-20), which is subsequently cleaved off to produce the mature form. The mature form contains an extracellular domain (aa 21-728), a transmembrane domain (aa 729-751), and a cytoplasmic domain (aa 752-798). Integrin beta-1 heterodimerizes with either alpha-1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11 or alpha-V to recognize different ligands. For example, alpha-1/beta-1 can recognize the proline hydroxylated sequence in collagen and alpha-2/beta-1 can serve as receptor for fibronectin. It is shown that beta-1 integrins recognize the sequence R-G-D in a wide array of ligands. Five different isoforms of integrin beta-1 have been described that are produced by alternative splicing. Isoform 1 is shown to be widely expressed while other isoforms are generally co-expressed with a more restricted distribution. Also, isoform 2 is reported to Interfere with isoform 1 resulting in a dominant negative effect on cell adhesion and migration.
